in non competitive inhibition , the inhibitor always binds to enzyme only when substrate is not present and in un competitive inhibition the enzyme will only attach to ES complex . so the kind of inhibitor , in each case has the property to sense whether ES complex is formed , and the presence of substrate right how this property ?

The sensing of this binding is based on the conformational or structural changes that enzyme or enzyme substrate complex undergo when inhibitor or substrate approaches them. In case of presence of enzyme molecule only, the inhibitor has binding site for the active site of enzyme. While in case of ES complex, the binding site for inhibitor becomes available only after enzyme binds with a substrate molecule.
